چکیده

In the present study, surface CD1d, which is involved in immune cell interactions, was assessed for effects on hematopoiesis. Mouse BM hematopoietic stem cells (HSCs) and hematopoietic progenitor cells (HPCs) express CD1d. The numbers and cycling status of HPCs in the BM and spleen of different strains of cd1d / mice were enhanced significantly, suggesting that CD1d is a negative regulator of HPCs. In support of this, CD1d was required for the SCF and Flt3 ligand synergistic enhancement of CSF induction of HPC colony formation and for HPC response to myelosuppressive chemokines. Colony formation by immature subsets of HPCs was greatly enhanced when normal, but not cd1d / , BM cells were pretreated with CD1dAbs in vitro. These effects required the full CD1d cytoplasmic tail. In contrast, long-term, but not short-term, repopulating HSC engraftment was impaired significantly, an effect that was minimally influenced by the presence of a truncated CD1d cytoplasmic tail. Pretreatment of normal BM cells with CD1d Abs greatly enhanced their engraftment of HSCs. The results of the present study implicate CD1d in a previously unrecognized regulatory role of normal and stressed hematopoiesis. (Blood. 2012;119(24):5731-5741)

بررسی اثر آگونیست -آدرنرژیکی ایزوپروترنول بر بیان miR-886-3p و miR-23a در سلولهای بنیادی مزانشیمی مغز استخوان انسان

Background and Objective: Mobilization of Hematopoietic Stem Cells (HSCs) for transplantation and the importance of -adrenergic signals in induction of this process have been well investigated. However, little is known about the role of -adrenergic signals in mobilization of HSCs and factors influenced by these signals. The Chemokine Stromal Derived Factor -1 (SDF-1) which is expressed by hum...

Pre-treatment with rapamycin protects hematopoiesis against radiation injury

Background: Protection of hematopoietic system has become a primary goal in the development of novel medical countermeasures against ionization radiation and radiotherapy. This study was to explore the role of rapamycin in normal tissues against radiation. Materials and Methods: Mice were pretreated with rapamycin by i.p. every other day for five times before 5 Gy or 8.5 Gy γ-ray whole bo...

Effect of daunorubicin drug with and without cimetidine on the nucleated cells of bone marrow of balb/c mouse

Introduction: Hematopoiesis is an on going process mammalian marrow system. A few cells from the nucleated cells of bone marrow are hematopoietic cells which include primary stem cells, precursor cells and progenitor cells. Primary stem cells and progenitor cells are able to produce colonies in culture medium (CFU-C) and irradiated mouse spleen (CFU-S). A hematopoietic cell is alive and act...

Gene Expression and Promoter Methylation Status of VHL, Runx-3, E-cadherin, P15 and P16 Genes During EPO-Mediated Erythroid Differentiation of CD34+ Hematopoietic Stem Cells

Background: VHL (von Hippel-Lindau), Runx-3 (Runt-related transcription factor 3), E-cadherin (Epithelial cadherin), P15 (INK4a, cyclin dependent kinase inhibitor), and P16 (INK4b) genes are essential in hematopoiesis. The aim of this study was to explore the correlation between gene expression and promoter methylation in CD34+ stem cells before and after differentiation to erythroid lineage. M...
